Please follow these steps while editing the Hotspot on the Chrome Extension:

1 - First, relocate it.
2 - Then, re-select it through the Custom CSS Selector option.
3 - Finally, check its z-index value.

Note: Ensure that the Page Targeting rules are valid and that your Hotspot is active under its settings.
